Evaluation of biological integration and inflammatory response to blood vessels produced by tissue engineering: experimental model in rabbit

Abstract

Introduction: The cardiovascular disease is the main cause of mortality in the western population and the Arterial Occlusive Disease (AOD), as a part of this group of diseases, is present in 5% of the adult population. As the disease advances, 25% of the symptomatic patients evolve to the amputation of the affected limb. Therefore, researches have been developed aiming to produce new arterial substitutes, being the cellular engineering a promising perspective. Goals: To synthesize blood vessels using scaffolds of rabbit's Inferior Vena Cava (IVC) and test its interaction with the receiver tissue and test inflammatory changes. Methods: The IVC will be obtained from 8 rabbits to descellularization or in natura veins. Each IVC will be fragmented in 2 segments of 2cm.The descellularized veins (DV) will be obtained through protocols of descellularization previously established, using Sodium Dodecyl sulfate 1% (SDS) and Sodium deoxycholate 2% (SD). 15 animals will be used to the experiment in vivo (tripled), being the product implanted in the interescapular dorsal area. Groups: 1-) in natura allogeneic vein; 2-) DV-SDS; 3-) DV- SD; 4-) DV-SDS + Mesenchymal stem cells of the adipose tissue (MSCat) autologous; 5-) DV-SD +(MSCat) autologous. The (MSCat) of the autologous receptors will be collected 21 days before the implant and expanded in vitro. Then, 60 days after, the animals will be sacrificed. The explants of DV/ in natura veins will be analyzed by histomorphological/immunohistochemistry. Peripheral blood will be collected in the pre operatory in 1d, 7d, 14d, 30d and 60d post operatory to dose the inflammatory cytokines. (AU)
