Abstract

Preeclampsia (PE) is a gestational specific syndrome characterized by a maladaptation state of the immunological tolerance, identified by oxidative stress and abnormal activation of the innate immune system. In plasma of pregnant women with PE there are elevated levels of molecular structures associated with stress and cell death, called damage-associated molecular patterns (DAMPs) such as uric acid, which appear to contribute directly to the pathogenesis of this disease. These DAMPs can activate monocytes switching them to the M1 profile. The imbalance between pro- and anti-inflammatory states in PE may be improved by administration of silibinin, a flavonoid with anti-inflammatory properties. This work aims to evaluate the immunomodulatory effect of silybinin on the expression of inflammatory markers and on the nuclear transcription factor B (NF-kB) signaling pathway in monocytes from pregnant women with PE and in THP-1 lineage of human monocytes activated with MSU.Monocytes from pregnant women with PE will be cultured with or without silibinin and THP-1 cells will be cultured with or without monosodium urate (MSU), and/or silibinin for different periods: a) 30 min for analysis of protein expression of IKB-± and NF-kBp65 by flow cytometry, b) 18 h for expression analysis of the surface markers TLR4, CD64, CD163 and CD14 and for determination of IL-1², IL-6, IL-8, IL-10, IL-12p70 and TNF-± by flow cytometry, and IL-23 and TGF-² concentrations by ELISA. The results will be analyzed using parametric or nonparametric tests with 5% of significance level. (AU)
