Abstract

In the early stages of the development of new drugs the study of in vitro metabolism is essential to understand the metabolic profile of the new chemical entity. The studies are traditionally carried out in solution, requiring a large number of tests to trace this profile, which results in the use of large volumes of microsomal fraction, cofactors and sample preparation time for further analysis. Thus, novel analytical strategies that minimize costs and increase test efficiency are of paramount importance for in vitro drug metabolism studies.In this scenario, the present project aims to develop a model of bioanalytical assays through the immobilization of microsomal fractions in capillary supports of fused silica and magnetic particles. Through the developed prototype the in vitro metabolism assays can be automated, thus simplifying the characterization of the metabolites produced. In addition, the test model meets the demands of green chemistry regarding the reduction of waste generated, low consumption of biological material, and reuse of the enzymatic system in various analyzes.
