Abstract

The present study aims to develop a liquid mouthwash formulation (ETF) containing 3% pomegranate peel extract, 0.2% sodium trimetaphosphate (TMP) and 100 ppm fluorine (F), as well as to evaluate its anticariogenic potential, irritant potential and cytotoxicity. The extract of the pomegranate peel will be obtained by alcoholic means and subjected to pharmacopoeial analysis by colorimetry of Folin Denis and Liquid Chromatography of High Efficiency. After obtaining the pomegranate extract, the ETF will be prepared, characterized and subjected to stability analysis. In situ evaluation of the anticariogenic potential will be performed in blocks of bovine tooth enamel coupled to palatine devices containing 4 enamel blocks that will undergo cariogenic challenge (sucrose 30% 6x / day). Volunteers (n = 12) will use the device for 7 days in 4 experimental phases testing the ETF and control groups. After the experimental period, the concentration of alkali-soluble carbohydrates (EPS), fluoride (F), calcium (Ca) and phosphorus (P) will be determined in the biofilm grown on the enamel block. The percentage of surface hardness loss, the hardness in longitudinal section, F, Ca and P present in the enamel will be determined. The irritation potential of the formulation will be determined by the hen's egg testing on the chorioallantoic membrane in four independent trials and will be compared to LISTERINE® Anticaries Zero Alcohol. The cytotoxicity test will be performed on primary cultures of rat fibroblast cells (L9F29), where cell viability will be assessed by the Alamar Blue assay. For statistical analysis, the most appropriate test for the distribution of data will be used, through Sigmaplot© software for Windows version 12.0, with significance at the 5% level. (AU)
