Abstract

Matrinxã (Brycon amazonicus) is a specie of great interest for a commercial fish culture due to its fast growth and excellent meat flavor. In view of this, studies are needed to improve the technique of artificial reproduction, such as the ideal proportion of spermatozoa and oocytes. Thus, the objective of the project will be to determine adequate inseminating dose for use in the artificial fertilization of Brycon amazonicus oocytes. Fourteen breeding animals will be used, eight males and six females, from the comercial fish farm. After selection, the fish will be submitted to hormonal induction, with two preparative dosages for 0.5mg and 5.0mg females of crude carp pituitary extract EBHc/kg body weight and single dose of 2.5mg/kg body weight for males. The gametes will be collected after 160 degree-hour (accumulated thermal units - ATU) consequent artificial fertilization to "dry" and later evaluated. Sperm analysis of concentration, survival, morphology and computerized motility analyzes will be performed in triplicate from the semen collection (semen pool). Computerized parameters of Motility Rate, Curvilinear velocity, Average path velocity, Straight line velocity, Straightness, Oscillation, Progression and Beat cross frequency will be evaluated using the free ImageJ / CASA plugin software, with configurations adapted to species. The collected oocytes will be distributed in a completely randomized experimental design, composed of five treatments and four replicates. The treatments will consist of the inseminating doses of 5,000, 50,000, 250,000, 500,000 and 1,000,000 mobile spermatozoa per oocyte. Will be consider as experimental unit the hatchery (1,5L) contained 1g of oocyte fertilized with the respective inseminant doses. After fertilization the eggs will be incubated until the moment of hatching. The results obtained from the rates of fertilization, hatching and normal larvae will be submitted to analysis of variance at 5% of significance and in case of effect will be applied regression analysis at the same level.
