Abstract

This project aims at developing innovative strategies for breast cancer chemoprevention based on nanotechnology and modern 3D models. This project is justified by the lack of strategies that are both well accepted and effective for chemoprevention in high-risk population. Herein, we have proposed the development of lipid-based elastic nanovesicles containing ethanol (20-45%) named ethosomes for topical administration and mammary tissue localization of the synthetic retinoid fenretinide. Microscopic techniques, light scattering and nano tracking analysis will be employed for evaluation of morphology, size distribution and zeta potential of ethosomes. To obtain a topical dosage form more easily administered, we propose to incorporate the ethosomes in a gel, and assess its rheological proprieties and stability. The influence of nanocarrier composition on the skin permeability, drug transport in/through the skin and irritation potential will be studied by consolidated methods. Subsequently, optimized formulations will be assessed for their antiproliferative activity in 2D and 3D breast cancer models.
