Abstract

In this project, we intend to study the immobilization of the enzyme pectinase (PEC) in Langmuir and Langmuir-Blodgett (LB) films of lipids, as well as to investigate their physicochemical properties and possible technological applications of the nanostructured systems produced. Characterizations of Langmuir monolayers include surface pressure-area isotherms, surface potential-area isotherms, Brewster angle microscopy, vibrational spectroscopy and rheological measurements; and for LB films the characterizations include UV-vis and infrared spectroscopies, atomic force microscopy and quartz crystal microbalance nanogravimetry, as well as viability of the LB films for pectin sensors by means of estimation of the enzymatic activity by colorimetric methods, and measurements of electrical capacitance to obtain analytical curves. In addition to the lipid-enzyme mixed nanostructured films, it is intended to further use other substances to determine whether they are capable of enhancing the molecular accommodation of the enzyme as well as improving the optical and electrical signal of the LB films. The candidate substances would be carbon nanotubes, metal nanoparticles, conductive polymers, and polysaccharides. The results will be evaluated from the perspective of possible implications in nanobiotechnology, inferred from the effects caused by carbon nanotubes, polysaccharides and conductive polymers in the artificial membranes, especially in the incorporation of enzymes.
