Analysis of the presence of virulence and resistance genes encoding antibiotic resistance and virulence determinants in S. marcescens and identification of vaccine candidates against S. marcescens using reverse vaccinology

Abstract

Multidrug resistant bacteria are emerging and spreading globally and even in different regions of the same country. In Brazil, it is no different and the studies about S. marcescens, a nocosomial pathogen with intrinsic resistance to several antibiotics classes and with a high potential of dissemination, are very scarce. The purpose of this study is to analyze the presence of virulence, porins and efflux pumps genes in S. marcescens, obtained from various clinical samples collected from patients admitted in ICU, by PCR analyses and posterior sequencing. Additionally, we will search for proteic targets that may be candidates to the production of vaccines, by bioinformatic analysis of the protein coding sequences from the genome of Serratia marcescens, and using the reverse vaccination technique (RV). Previous studies of our research group found that the majority of S. marcescens, used in this study, are multi-drug resistant (MDR), with a high level of ²-lactam, aminoglycosides, quinolone, tigecycline and colistin resistance. Initial studies are being conducted to verify the presence of ²-lactam and other antibiotic resistance genes. Therefore, the proposed study will extend our knowledge on pathogenic S. marcescens in Brazil, it will allow a comprehensive understanding of it's pathogenicity and, in the future, may contribute for the development of novel molecular diagnostic methods, new drugs, vaccines and, consequently in the control and prevention of nosocomial infections. Considering that the antibiotic resistance is one of the biggest threats to global health, our findings will also be useful to predict, nationally and internationally, the emergence of antibiotic resistant bacterial strains, to guide intervention strategies and to support new and better treatment plans.
