Abstract

Introduction: Leishmaniasis is one of the seven most important tropical diseases, representing a worldwide health problem with several clinical manifestations and can progress to death. In particular, photodynamic therapy has shown promising effects as a treatment of cutaneous Leishmaniasis, but with a need for standardization of treatment and greater investigation of the mechanisms of action. Objective: To analyze the effects of photodynamic therapy with methylene blue and red LED on promastigotes of L. amazonensis and L. braziliensis. Methods: Cultures of L. braziliensis and L. amazonenses will be performed, and for infection, J774 macrophages, PDT with methylene blue and red LED will be performed. Cell viability will be analyzed by mitochondrial activity with MTT assay, nitrite determination and nitrate will be performed with Griess method and morphology by ultrastructural analysis with transmission electron microscopy.
