Abstract The project aims to generate immortalized PDAC cell lines resistant to gemcitabine, and the molecular characterization of this resistance. Such lines will be generated by prolonged treatments under sub-lethal concentrations of the drug and will have their resistance evaluated by determining the expression of resistance marker genes already described in the literature by RT-qPCR. In addition to widely studied PDAC lines, a line derived from PDAC xenotransplantation will also be used, which more reliably recapitulates the patient's tumor microenvironment. The results obtained in this project will be the basis for a project under development in the Laboratory of Integrated Research in Cancer of the IQ-USP, which aims to perform the scan of components of the epigenetic machinery functionally associated with chemoresistance and sensitivity to gemcitabine in cancer of pancreas. | |
