Abstract

Considering the accelerated pace of new cases emergence of tuberculosis (TB), about 10 million per year, the high rates of resistance to main classes of antimicrobials, and taking into account that TB occupies the first place in the ranking of causes of death by an infectious agent, becomes evident the need to control and combat this disease which poses a serious threat to global public health. To do so, we must face the challenge of developing new active agents against TB. In this work, the metal complex, tris (1,10-phenanthroline)iron(II), ([Fe(Phen)3]2+), which has never before been described as a drug, will continue to be the object of study since it presented itself as a good candidate in previous tests against TB. In order to contribute to the research of new drugs that are effective and easily accessible for treatment, our objective will be to search for the mechanism of action of the complex ([Fe(Phen)3]2+) and its possible effects against Mycobacterium tuberculosis (MTB), through methodologies that allow the isolation and genomic sequencing of spontaneously resistant mutants. In addition, the complex will be incorporated into a microemulsified nanostructured lipid system, in order to increase stability and improve oral bioavailability, and its in vitro permeability will be assessed by the Caco-2 cell monolayer assay, the metal will be quantified by atomic emission spectrometry with inductively coupled plasma (ICP-OES) and the permeability of nanostructure confirmed by Confocal Laser Scanning Microscopy (CLSM).
