Cytotoxicity and inducing effect of mineralization of flavonoids on osteoblastic cells

Abstract

Dental bone loss and the formation of periapical lesions arise as a consequence of imbalance of bone homeostasis. Osteoblasts, together with osteoclasts and osteocytes, act in bone formation and resorption. Several markers of bone formation are produced by active osteoblasts and reflect different aspects of osteoblastic differentiation and bone remodeling. Thus, many authors have explored the use of phytotherapics in order to obtain new compounds with therapeutic properties, such as flavonoids, and also stimulate bone neoformation and periapical region repair. The objective of this study will be to evaluate in vitro the cytotoxicity and inducing effect of flavonoid mineralization on human osteoblastic cells. For this, osteoblastic cells of the Saos-2 lineage will be exposed to the following flavonoids: quercetin, myricetin and its derivatives taxifolin, isoquercitrin, rutin, amelopsin and EGCG alone or combined and evaluated the cytotoxic effect, alkaline phosphatase activity, mineralization nodules and the gene expression of mineralization markers on osteoblastic cells such as collagen-1 (COL-1), osteocalcin (OCN) and osteopontin (OPN). The quantification of the protein with highest gene expression (COL-1, OCN or OPN) will be also evaluated by ELISA assays. In addition, it will be possible to determine the structure-activity relationship based on the chemical differences between flavonoids and results obtained in the biological assays that may direct future work with the flavonoids. With this study, we intend to propose an intracanal medication with natural flavonoids which present therapeutic properties based on the in vitro results, aiming at a future clinical application.
