Proteomic analysis of acquired pellicle dental enamel: effect of cystatin, staterin and hemoglobin against dental erosion

The presence of acquired pellicle enamel (PAE) is one of the protective factors against tooth erosion. Among the acquired pellicle proteins with potential to bind to enamel and acid-resistant properties, cystatin, hemoglobin (Hb) and staterin were recently identified by our group. Modification of the protein profile of the acquired pellicle by incorporating some proteins may affect its ability to protect against tooth erosion. Therefore this modification seems to be promising for the control of dental erosion. The present work aims to evaluate the impact of the modification of PAE with acid-resistant proteins on their proteomic profile. Specifically, the objective will be to evaluate the effect of CaneCPI-5, staterin-derived peptide (Stn15pSpS) and hemoglobin, alone or in combination, on the proteomic profile of the in vivo formed EAP aiming ultimately at preventing erosion. A cross-blind and triple-blind study will be conducted, consisting of 5 parallel phases, corresponding to the treatments with the isolated proteins, in combination or with deionized water (negative control). At each stage, after prophylaxis, 2 volunteers will rinse for 1 min with each of the 4 protein solutions or deionized water. After the mouthwash, the SAP will be formed for 3 min or 2 hours. PAE collection will be performed with the aid of an electrode filter paper and the samples will be stored at -80 ºC. Subsequently, quantitative marker-free proteomic analysis will be performed (Xevo Q-TOF, Waters). PLGS software will be used to detect changes in protein expression between different groups.
