Evaluation of acute toxicity and gastroprotection activity of anthocyanin cyanidine in female rodents

Abstract

The interest in anthocyanins has increased significantly over the time in a number of areas (especially in the food and pharmaceutical industries), and the numerous pharmacological studies confirm the efficiency of these compounds present in several fruits, as a result of their different benefits in the most different diseases. In fact, anthocyanins presents in the diet have been considered as responsible for reducing the risk of diseases associated with its antioxidant and anti-inflammatory properties. The present project aims to evaluate anthocyanin cyanidin against acute toxicity and gastroprotective activity in female rodents. Cyanidin presents promising gastroprotective activity in male mice (FAPESP process 2016 / 20600-0). In order to investigate their safety, an acute toxicity test will be performed, as indicated by the OECD, in which female mice will be treated with a single dose of cyanidin, having several parameters compared to the vehicle group (histological analysis of the heart, kidneys and liver, analysis of plasma levels of AST, gamma-GT, AST, urea, creatinine, glucose). Additionally, aware of the differences between males and females, the absolute ethanol induction model will be performed in females to compare the activity in the same doses already evaluated in males. Different studies indicate that the pharmacological response differs in males and females, thus, this work aims to analyze if cyanidin also promotes gastroprotection in females, analyzing macroscopic parameters of gastric lesions, antioxidants (SOD, CAT, GSH), inflammatory (MPO) and morphological (histology). The results obtained will provide data on the acute toxicity and gastroprotective activity of cyanidin, as well as scientific subsidies for the future incorporation of these compounds as a therapeutic option in the different diseases of the gastrointestinal tract.
