Abstract

Many clinical studies demonstrate the impact of smoking on increased bone tissue fragility as well as on the development and progression of diseases that lead to loss of cartilage tissue, such as osteoarthritis. However, most of these studies are restricted to the analysis of imaging tests and symptoms of patients, which hinders a further understanding of the pathophysiological mechanisms involved in the repair process of these tissues after the established injury. Thus, to advance the understanding of the role of different cell types, chemokines and growth factors involved in the process of cartilage wear in osteoarthritis patients, samples of this type of tissue will be collected from smokers and non-smokers submitted to total arthroplasty. hip or knee. For this purpose, we will evaluate the main cellular type constituting the cartilage (chondrocytes), the chemokines and growth factors involved in the cartilage repair process and the tissue histoarchitecture. In this project, the patients will be divided into three experimental groups: Smokers (non-quit smoking patients before surgery), elective smokers (smokers who quit smoking at least 6 weeks before surgery) and non-smokers (patients who have never smoked before surgery). smoked). In cartilage tissue homogenate samples, chemokines directly involved in tissue repair metabolism such as RANKL, OPG, IL-1, IL-6, TNF-alpha will be evaluated by ELISA. Specific immunohistochemical staining will be done on cartilaginous tissue for the quantitation of chondrocytes and collagen type II.
