Abstract

The translation of mitochondrially encoded mRNAs is done by mitoribosomes. Mitochondrial translation involves several factors, including translation activators, which bind to a site in the 5 'UTR region of the mitochondrial mRNA and a site in the coding region. The factors Pet309, Pet111 and Pet494 have already been observed for the COX1, COX2 and COX3 genes, respectively. Several uncharacterized proteins in the MIOREX expression system encoded by the MRX genes have been discovered, and recent studies have revealed the role of some of these proteins in mitochondrial translation. Mrx5p, for example, inhibits the translation of the ATP6 / 8 bicistronic mRNA, Mrx15p interacts with the 54S subunit of the mitoribosome, and Mrx6p regulates the mtDNA copy number, while eleven Mrx proteins remain uncharacterized. Therefore, this project aims to characterize such Mrx factors in an attempt to elucidate regulatory mechanisms of mitochondrial translation. (AU)
