Abstract Oropouche virus (OROV) is an arbovirus present in the Amazon region and Central America. Oropouche fever has affected more than half million people in the last 50 years, causing fever, joint pain, headache and myalgia. Like other arboviruses, many cases are not diagnosed correctly and there is no specific treatment for the disease. The use of high throughput screening for drug discovery and repurposing of FDA human approved drugs has been shown as an alternative for new treatments, in behalf of high costs and the long time to approve new drugs. The present project aims to stablish and optimize an HTS assay for phenotypic studies using VERO cells infected with OROV, and to select compounds with potential cytoprotection against the infection. Preliminary Results: A stock of OROV was produced from the original strain BeAn 19991 for the HTS assay. In this screening campaign, it was used more than 1000 compounds from the National Institutes of Health Clinical Compound Collection (NIH-NCC) approved for human use. Of those, 2% of the compounds showed a decrease of the death of OROV-infected cells by at least 50%. (AU) | |
