Determination of ideal conditions for in vitro mucoadhesion assays in porcine buccal mucosa

Abstract

Mucoadhesive drug delivery systems have been extensively investigated for the topical administration of drugs on mucosa, especially the oral mucosa. These systems offer the advantage of increasing the contact time of the drug with the buccal mucosa, which is highly vascularized, and thus, it can increase the drug bioavailability at the action site, avoiding the effects of first-pass hepatic metabolism. Thus, such systems have enabled both the local treatment of pain, inflammatory conditions and oro-dental infections, as well as the treatment of systemic pathologies by buccal route. In this context, the assessment of mucoadhesion of topical formulations is an in vitro methodology that has been widely used using several methods, especially the evaluation of the detachment force by texture analyzer. However, this methodology has several instrumental variables that can affect the results making it difficult to compare different studies. Therefore, the objective of the present project is to determine the operational condition that results in greater mucoadhesive capacity in vitro of a commercial buccal formulation in fresh pig oral mucosa, as well as the determination of reference values for future studies. In this way, a statistical plan 2^3 composed of 11 tests (eight experimental points and 3 central points) will be carried out in order to evaluate the influence of the variation of operational conditions, such as probe speed, contact time and contact force, in the mucoadhesive properties called peak of mucoadhesive force and mucoadhesion work. Such properties will be measured by the equipment Texture Analyser TA.XT. Plus Stable Micro Systems and calculated using Exponent software. Thus, we intend to optimize this method of mucoadhesion analysis and establish reference values for the mucoadhesion parameters in order to contribute to the development of new mucoadhesive drug delivery systems for topical use in the oral mucosa for the treatment of various conditions, especially in Dentistry.
