Comparison of the taxonomic and transcriptomic profile of subgingival biofilm associated with periodontitis and peri-implantitis

Abstract

The objective of this cross-sectional, control case study will be to evaluate and compare the microbial profile (taxonomic and transcriptomically) and inflammatory cytokines of peri-implantitis and periodontitis sites, in addition to determining how metal wear can impact the relationship between these variables. Forty participants diagnosed with Grade B / C, stage 3-4 periodontitis or with peri-implantitis will be selected (n = 20 in each group) to participate in the study. They will be allocated into two groups, according to the diagnosis of their disease (periodontitis group and peri-implantitis group). Initial clinical parameters will be evaluated through a millimeter-gauged probe of patients with periodontitis and peri-implantitis. Biofilm collection will then be carried out using sterile endodontic paper points from a tooth / implant from each patient of both groups for subsequent DNA extraction (Qiagen MiniAmp Kit), sequencing (Illumina MiSeq) and data evaluation for taxonomic analysis. Metatranscriptomic analysis will also be performed from the same biofilm samples. There will be the collection of gingival fluid through sterile endodontic paper points to analyze the inflammatory markers of each group (Luminex / MAGpix equipment) and, from the crevicular fluid, the concentration of Titanium present will be analyzed.
