Role of MMPS/TIMPS expression on peri-implant repair using an in vitro model

Abstract

Incidence of failure of oral osseointegrated implants can be related to increased expression of matrix metalloproteinases (MMPs). These enzymes act as collagenases, remodeling peri-implant tissue, however, their overexpression may result in exacerbated extracellular matrix degradation, delaying or impairing healing. MMPs are endogenously controlled by proteins called tissue inhibitors of matrix metalloproteinase (TIMPs). Therefore, he balance between synthesis of MMPs and TIMPs guaranties the tissue homeostasis and also tissue repair and remodeling processes. The aim of this investigation will be to determine the role of expression of MMPs and TIMPs for gingival fibroblasts and osteoblasts seeded onto titanium discs and submitted to inflammatory challenge. Titanium discs will be individually placed in cell culture plates and cells will be seeded onto these discs. After 24 hours, inflammatory stimuli will be added to culture medium (TNF-alpha- 100ng/ml) and will be maintained for additional 24 hours. Then, synthesis of MMP-2 and -9, as also TIMPs 1 and 2will be determined by ELISA. Data will be statistically analyzed considering distribution, homogeneity and homocedasticity.
