Abstract

The postdoctoral scholar will achieve these objectives: *Collect air samples through filtering during height profiling performed in balloon flights in different locations in Brazil at different seasons (winter/summer) and during harvesting; *Collect samples from soil, coffee and grass leaves in order to detect INA (ice nucleation activity); *Freezing tests using PORTABINA device; *Calculate and/or estimate IN microbiota flux from the ground; *Test half filters (ground and balloon) for IN activity using Conen et al. methodology (Atmos. Meas. Tech., 5, 321-327, 2012), on temperatures between -2oC and -10oC. The other half will be used for total DNA extraction and 16s diversity characterization; *On the filters with positive IN activity, proceed to isolation on generic medium and on medium specific for Pseudomonas, confirmation and characterization of IN activity, and 16s identification of isolates with positive results. **These steps have the scientific goals of: *Assessing the mechanism for the influence from bio-aerosols on the simulated clouds, identifying the nucleation and microphysical growth pathways in clouds, using models. *Quantify the INA impacts on clouds from the various species of bio-aerosols by doing sensitivity tests.
