Influence of MitoQ10 supplementation on cardiac remodeling after experimental myocardial infarction

Abstract

After Myocardial Infarction (MI), changes occurs in the ventricular architecture, which have been studied in recent years as Cardiac Remodeling (CR). Several factors are associated with CR after MI and, among them, changes in energy metabolism, oxidative stress, inflammation and, more recently, the metabolomics and intestinal microbiota are also been studied. Mitochondria are one of the main sites for the generation of Reactive Oxygen Species (ROS) in cells. However, the effectiveness of conventional antioxidants is limited due to the difficulty of being delivered to mitochondria in situ. In this sense, a ubiquinone has recently been developed and adapted with the mitochondrial target, being called MitoQ10. The aim of this study is to evaluate the influence of the antioxidant MitoQ10 on the CR process after experimental IM. Male Wistar rats will be submitted to experimental infarction or to sham surgery. After the initial echocardiographic study, the animals will be allocated into 4 groups: 1) Sham group - animals not submitted to IM receiving standard chow (SC - n = 20); 2) Sham group fed a standard show supplemented with the MitoQ at a dose of 10 micromol/kg (SMQ group - n = 20); 3) infarcted group (animals submitted to experimental infarction) fed standard chow (IC group - n = 20); 4) infarcted group fed a standard chow associated with MitoQ supplementation at 10 micromol/kg (IMQ group - n = 20). After three months morphological, functional and biochemical studies will be performed. In cardiac tissue morphometry, microRNA, oxidative stress, energy metabolism and protein expression will be analyzed. In the serum of the animals will be performed metabolomics analysis, and feces will be collected before and after the supplementation to evaluate the intestinal microbiota. Values will be presented as mean ± standard deviation and comparisons will be made by two-way ANOVA test, with a significance level of 5%. (AU)
