Abstract HNSCC is the sixth most common type of cancer in the world and affects anatomic regions, which include the oral cavity, pharynx and larynx. Its main etiological factors are smoking, alcohol consumption and human papillomavirus (HPV) infection. The metabolic profile of HNSCCs is heterogeneous and related to mutations in several tumor suppressor genes and oncogenes. Altered metabolic patterns include increased levels of lipids in biofluids, a finding confirmed by previous data from our group. The aims of this project are (1) to investigate the spatial distribution of lipids directly in tissue sections of tumor and surgical margin samples from patients with oral squamous cell carcinoma (OSCC) using mass spectrometry imaging (MSI) technique; (2) to characterize the plasma lipid profile in patients with OSCC and controls by capillary electrophoresis technique coupled to mass spectrometry (CE-MS); (3) to evaluate whether changes in the plasma and tumor lipid profile are correlated with clinical and histopathological characteristics; (4) to investigate the correlation between the expression of choline-metabolizing enzymes in tumor samples and the level of phospholipids in plasma of patients with OSCC; (5) to investigate the correlation between the expression of inflammatory mediators in tumor samples and the level of phospholipids in plasma of patients with OSCC; (6) to evaluate the role of candidate biomarkers associated with lipid metabolism in OSCC using functional in vitro assays and their potential use in non-invasive approaches.
