Abstract

Tissue engineering and bone tissue regeneration have become a very important areas for the development of alternative functional materials to restore, maintain or improve the function of damaged tissue. Presently, after the technological advance in science, the development and improvement of biomaterials have become necessary to evolve biomaterials that will contribute to the population's quality of life. Thus, the aim of this study will be to evaluate the influence of calcium aluminate and zirconia additives in polymethylmethacrylate (PMMA) bone cement on in vitro osteogenesis with cells obtained from rat femurs. After production, the surface morphology of the samples will be analyzed by scanning electron microscopy (SEM), and subjected to wettability tests using the contact angle technique, qualitative analysis of the chemical composition of the sample's chemical elements, and x-ray diffractometry. After characterization, the samples will be submitted to in vitro biological tests using rat femurs mesenchymal cells. The cells will be isolated and plated with the samples to assess the influence of biomaterials on osteoblast cell activity and differentiation, through cytotoxicity tests, assessment of total protein content, alkaline phosphatase activity, analysis of cell morphology, genotoxicity, and formation of mineralization nodules. We will also evaluate the production of TNF-± and IL-6 cytokines. All biological tests will be carried out in three independent experiments. The data will be statistically analyzed with the Graph Pad Prism software, initially through the normality test, aiming to apply the appropriate statistical test. The significance level adopted will be the conventional value of 5%.(AU)
