Abstract

Breast cancer is characterized by cell abnormality, genetic instability and disordered growth, representing about 25% of cancers that harm women's health, with high mortality rates resulting from this pathology. In addition, the treatments available today are chemotherapy, radiotherapy, surgery, and, more recently, immunotherapy, whose effects are not always satisfactory and may present recurrence, metastasis, aesthetic and psychological damage. On the other hand, photodynamic therapy (PDT) stands out as an alternative, as it presents a satisfactory response. The project seeks to evaluate the cellular mechanisms triggered by PDT in breast cancer cells (MCF7 lineage) and normal breast epithelial cells (MCF10), in 3D culture. For the study, 4 groups will be divided for each strain: Control, untreated cells; AlPcS4, cells incubated with the photosensitizer; Light, cells irradiated with a wavelength of 660nm and TFD, cells incubated with photosensitizer and irradiated. For irradiation, the Biopdi/IRRAD-LED (660 nm) device will be used, with an irradiance of 25 mW/cm² and an exposure time of 3 min and 20 s. Thus, cell viability will be evaluated through flow cytometry, by labeling with propidium iodide, detection of heat shock proteins HSP27, HSP70, GRP78, and HSP90, and intracellular glycoproteins. The analysis of surface glycoproteins will be performed by Western Blotting and for evaluation of the phagocytosis process, the breast strains, after the procedures, will be incubated with macrophages (RAW 264.7).(AU)
