Abstract

This study aims to investigate the local action of Rubus coreanus through the functionalization of the surface of the implant to be installed in the tibiae of female rats with estrogen deficiency. Forty-eight young adult female Wistar rats will be used. The experimental groups will be SHAM CONV, SHAM RC 200, SHAM RC 400, OVX CONV, OVX RC 200, AND OVX RC 400. The OVX animals will be submitted to bilateral ovariectomy, while SHAM animals will undergo sham surgery. After 30 days, all animals will undergo implant installation surgery in the tibial metaphysis. The implants will have the surface treated with Rubus coreanus with different concentrations; having as negative control the implants with the conventional surface, from the factory. The animal's euthanasia will occur 28 days after installation of the implants and tibias will be destined for the following analyses: Real-time PCR (expression of ALP, OCN and TRAP genes), while the contralateral side will be performed computerized microtomography for morphometric evaluation of bone formed at around the installed implants. Subsequently, these same samples will be submitted to routine laboratory processing for decalcified cuts, embedding in paraffin, and for immunohistochemical evaluations in order to detect the presence of Alkaline Phosphatase, Osteocalcin and TRAP proteins. Quantitative data will be submitted to the homoscedasticity test to select the appropriate statistical test (parametric or non-parametric), with a significance level of 5%. (AU)
