Abstract

When dealing with biopharmaceuticals, suitable product quality, minimal rejection rates, as well as high yield and process productivity, will largely depend on rigorous control of critical parameters throughout the manufacturing stages. This control is made possible by monitoring the process. To target this goal, analytical techniques are often applied to monitor biochemical parameters and it is preferable that they are affordable, providing accurate and fast measurements. Thus, the use of spectroscopic information associated with linear and non-linear correlation techniques has been shown to be an advantageous alternative for online and offline bioprocesses monitoring. These techniques are non-destructive, non-invasive, and provide both the rapid chemical analysis of molecules of interest and simultaneous monitoring of multiple compounds. The modeling of the data obtained through these techniques can show the level of correlation between multiple process parameters and allow predictions about them to be made. Thus, the present project aims to compare the predictive capacity of four correlation techniques: partial least squares, principal components regression, artificial neural network, and support vector machines. The models derived from these tools will allow the connection of process variables quantified by classical methods and spectral data from off-line monitoring of the production of rabies virus-like particles by Raman spectroscopy in samples without any processing (with cells) and samples of filtered supernatants (without cells). The fitted models from the four techniques will make it possible to estimate nutrients, metabolites, cell density, and concentration of viable cells in the samples, favoring the monitoring of the next stages of bioprocess development.
