Abstract

Apical periodontitis (AP) is characterized by pulp necrosis leading to periapical bone degradation, consequence of bacterial colonization of root canals. It is a serious public health problem, because there is a bidirectional relationship between oral infections and systemic health. Endodontic medicine seeks to investigate alternative therapy options against this disease, in which herbal medicine has a great potential on the rise. Yerba mate (Ilex paraguariensis) has a large source of health-promoting phenolics, as well as anti-inflammatory, antimicrobial and antioxidant effects that have been described in the literature, and also beneficial effects on bone biology. The stem cell used will be cells of dental origin from human exfoliated deciduous teeth (SHED), for their high proliferation capacity and for being osteoinductive. Objective: To evaluate in vitro, using SHED cells the extract of yerba mate (YM) on the cytokine profile present in apical periodontitis and osteoclastogenesis. Methods: YM tea powder at concentrations of 10, 20 and 50 ¼g/mL will be used. Cellular viability analysis will investigate the cellular cytotoxicity of YM in different concentrations. In osteoclastogenesis analysis investigate the effect of YM on bone repair and absorption and finally investigate the action of YM on umportant AP mediators such as IL-1a, IL-6, IL-10, OPG and RANLK. Analysis of results: The results will be submitted to statistical tests specific for each case, and a significance level of 5% will be adopted. (AU)
