Abstract

Diabetic retinopathy (DR) is one of the most serious and common complications of diabetes and considered the leading cause of acquired blindness. During the development of DR, there are morphological changes in the retina and activation of Müller glial cells that contribute to the inflammatory process, which may lead to retinal degeneration. For relevance to this project and the lack of safe long-term treatment, we highlight Annexin A1 (AnxA1), an anti-inflammatory protein capable of regulating neuroinflammation and neurodegeneration. Thus, it is essential to study the action of AnxA1 in streptozotocin (STZ)-induced DR in wild-type (WT) and knockout (AnxA1-/-) mice. We will use the hyperglycemia model in human Müller glial cells (MIO-M1) treated with Ac2-26 or Boc2 to evaluate gliosis and release of pro-inflammatory mediators. Bringing AnxA1 closer to the ophthalmology clinic, we will investigate this protein and inflammatory cytokines in the vitreous and blood plasma of patients with proliferative DR, with the aim of proposing AnxA1 as a biomarker and correlating it with inflammation, phase and time of disease development. The results obtained in this project will allow to expand the understanding of the role of AnxA1 protein in the progression of this eye disease, in order to extrapolate this knowledge to translational studies and benefit individuals affected by DR.
