Effect of glycolic acid on the bond strength longevity of bioceramic endodontic sealer to root dentin

Abstract

The aim of this study will be to evaluate the influence of glycolic acid solution as a final irrigant on the longevity of the bond strength of bioceramic and resin endodontic sealers to root dentin. Forty-eight single-rooted human teeth will have their crowns removed and the root portion subjected to biomechanical preparation with the Reciproc system (R50) and irrigation with 2.5% sodium hypochlorite. The specimens will be randomly distributed into 4 experimental groups according to the final irrigation protocol and endodontic sealer (n=12): Group I - EDTA (17%) + NaOCl + resin based sealer; Group II - Glycolic Acid (10%) + NaOCl + resin baser sealer; Group III - EDTA (17%) + NaOCl + bioceramic sealer; Group IV - Glycolic Acid (10%) + NaOCl + bioceramic sealer. After the final irrigation protocol, the canals will be dried and filled with Bio-C Sealer, bioceramic endodontic sealer, or AH Plus, resin based sealer, according to the experimental group. In 2 samples of each group dye will be added to the sealers during manipulation: rhodamine B 0.1% will be used for the resin based sealer, and 0.3% calcein-AM for the bioceramic sealer. After 3 times the sealers setting time, the specimens will be cross-sectioned into slices of 1 mm (± 0.1 mm) thick, obtaining 3 slices from each third of the root. The first slice of each third (n=10) will be used for the immediate push-out test followed by the analysis of the failure pattern; the second slice of each third (n=10) destined to the push-out test after 6 months followed by the failure pattern analysis; the third slice of each third will be used to analyze the quality of the adhesive interface in laser confocal microscopy (CLM) (n=5). In samples in which the sealers receives dye, 2 slices of each third will be prepared for the analysis of the intratubular penetration of the sealers by fluorescence laser scanning confocal microscopy. The data obtained will be analyzed with ANOVA, chi-square and Kruskal-Wallis test.
