ARTEMISININ AND CISPLATIN EFFECTS ON LEUKOPLAKIA PROGRESSION INTO ORAL SQUAMOUS CELL CARCINOMA IN A PRE CLINICAL MODEL

Abstract

Alcohol, tobacco and the presence of potentially malignant disorders (PMD), as leukoplakia, are important risks for the development of a squamous cell carcinoma (SCC). Artemisinin (ART) and cisplatin (CIS) have evidence that they may be useful to prevent the progression of leukoplakia into a SCC, thus, we aim to evaluate if interventions with ART and CIS are effective in the treatment of mouth leukoplakia in mice, preventing the progression to a SCC. 50 C57BL/6J mice wil, be divide in 4 groups, pure control, control, CIS and ART. 4NQO will be administered in the water to induce leukoplakia in the groups: control, CIS and ART. The control group will receive DMSO, CIS will receive a dose of 5 mg/kg of the chemotherapeutic and the ART group will receive 85mg/kg, all intraperitoneally. In the periods of 8, 12 and 16 weeks, the animals will be euthanized, the tongues will be collected, fixed, processed, stained with hematoxylin and eosin and evaluated by conventional light microscopy. The absence or presence of hyperkeratosis, degree of dysplasia and the presence or absence of SCC will be evaluated, the data will be statistically evaluated by the normality test and the appropriate statistical test will be applied with a significance level of 5%.
