Evaluation of adjuvant therapies of systemic administration in the overlying gingival tissue in post-extraction areas in senile rats treated with zoledronate at an oncological dose

Abstract

Drug-related osteonecrosis of the jaws (MRONJ) is characterized by aseptic necrosis of bone tissue due to the use of antiresorptive or antiangiogenic drugs. Its etiopathogenesis is still unknown, making it more difficult to choose an effective protocol for its treatment and cure. Adjuvant therapies can play a key role in controlling these cases, such as therapy with pentoxifylline and tocopherol. In addition, systemic ozone therapy has been widely used in dentistry due to its antimicrobial properties and its optimization in soft tissue repair due to the expressive increase in the contribution of growth factors promoting neovascularization. Combining these two points, the objective of this work will be to evaluate the potential use of systemic therapies, such as pentoxifylline (PENTO) and tocopherol (TOCO), associated or not, as well as systemic ozone therapy (OZ) for the prevention, control or even same cure of MRONJ, therefore, 27 female Wistar rats, 18 months old, where 15 will undergo bone tissue characterization, the others will be divided into 6 experimental groups, each group with 2 rats and each rat with 2 surgical sites n=4; SAL group, where the rats will be submitted to applications of 0.9% sodium chloride solution and the ZOL group will receive application of zoledronate 100¼g/kg, both every 3 days for 7 weeks; groups ZOL+PENTO, ZOL+TOCO, ZOL+TOCO+PENTO, group ZOL+OZ, all animals will be submitted to the installation of tethering around the lower first molars and will receive the respective therapies over three weeks, where to perform - There will be the extraction of these elements, following up with the applications for 28 days after surgery, where euthanasia will occur for clinical analysis, and collection of overlying gingival tissue for histological/histometric analysis. Quantitative data will be submitted to the normality curve to determine the most appropriate test (p<0.05).
