Abstract The Bothrops genus is the main cause of snakebite accidents with envenomation in Brazil, and the venoms of these snakes cause severe local and systemic effects, due to the high concentration of different enzymes, including proteases and phospholipases A2. At the site of bite, tissue damage is an immense concern, as connective tissue and skin cells come into direct contact with toxins. The fibroblasts, important components of connective tissue, act in the healing of inflammatory and traumatic injuries, thus the identification of the alterations caused by the effect of toxins is essential to understand the molecular pathways related to pathological changes. Thus, the aim of this project is to molecularly characterize the effects generated in human skin-derived fibroblasts (Hs68-ATCC cell line) after exposure to B. jararaca venom. To this end, after the assessment of cell viability and proliferation, the secretome will be collected and proteomic and peptidomic analyses will be performed using approaches of mass spectrometry.Key-words: Bothrops jararaca; fibroblasts; secretome; proteomic; peptidomic; mass spectrometry. | |
