Abstract

This research project aims to evaluate the extraction, separation and bidimensionalanalysis of phenolic compounds from agro-industry residues with antimicrobialpotential against Staphylococcus aureus for application in curatives. Initially, fastanalysis methods will be developed for the identification and quantification by ultrahigh pressure liquid chromatography (UHPLC) of the compounds present in samples ofdifferent agro-industry residues. The fractionation of compounds present in thesesamples will be studied through the on-line coupling of pressurized liquid extraction(PLE) in gradient with solid phase extraction (SPE) (PLEgrad-SPE) with real-timemonitoring. Real-time monitoring of the extraction/separation process will be carriedout through the orthogonal coupling PLEgrad-SPE x UHPLC and the simultaneouselaboration of kinetic extraction curves of several compounds present simultaneously.The compounds present will be identified and two-dimensional profiles of the chemicalcomposition of the fractions will be prepared obtained through orthogonal couplingPLEgrad-SPE x UHPLC. Fractionated extracts of samples will be obtained to carry outthe evaluation of antimicrobial activity against Staphylococcus aureus and selection offractions with greater activity. The best way to apply the selected phenolic compoundsin films for application in curtives will be evaluated, for which: (i) the necessity ofencapsulating the bioactives will be tested; (ii) films containing the bioactives will bedeveloped, testing different options: chitosan, gelatin and/or starch films, and (iii) thefilms will finally be characterized for later application in curatives/dressings.
