Abstract

The literature reports that the consumption of western diet can trigger damage to skeletal muscles by several metabolic pathways. To preserve skeletal muscle tissue, the use of bioactive compounds has been tested. It is described that the Gamma-Oryzanol (³Oz ) has great influence on the mechanisms involved in skeletal muscle metabolism. Even though these reports are already established, studies with ³Oz addressing ³ broad aspect investigating all metabolic pathways in skeletal muscle of animals submitted to the Western diet have not yet been performed. Therefore, the aim of this study is to improve the therapeutic capacity of ³Oz in metabolic alterations present in muscle s with oxidative capacity (Soleus) and glycolytic (Extensor digitorum longus muscle (EDL)) of Wistar rats submitted to western diet consumption. Wistar rats (n=32) will be randomly distributed into two groups to receive, ad libitum, control diet (C, n= 16 animals) and diet rich in sugar and fat plus 25% sucrose in drinking water (HSF, n= 16) for 20 weeks to induce obesity. After this period the animals will be redistributed into 4 groups, receiving, ad libitum, control diet (C, n=8 animals), control diet + Gamma-oryzanol (C + ³Oz, n=8 animals), diet rich in sugar and fat (HSF, n=8 animals) and diet rich in sugar and fat + Gamma-oryzanol (HSF + ³Oz, n=8 animals) for 10 weeks. Gamma-oryzanol will be increased to 0.5% in the feed. The consumption of the animals will be measured daily to evaluate the caloric intake and the weight measured weekly. The index of adiposity, glucose, triglycerides and insulin will be evaluated at the end of the study. Insulin resistance will be calculated by calculating HOMA-IR. In muscle tissue, oil and EDL, inflammatory markers (IL-6, TNF-±), oxidative stress (Malondialdehyde, protein carbonylation and antioxidant enzymes) and ectopic accumulation of fat via muscle triglycerides will be measured. The Western Blot technique will also be measured in the Soleus and EDL muscles, the GLUT-4. Skeletal muscle tissue metabolites will be identified and quantified through metabolomics.
