Effect of applying copper-doped 45s5 bioglass to gutta-percha and its interaction with bioceramic sealers

Abstract

Gutta-percha (GP) associated with endodontic sealers is the solid material of choice for filling root canals. However, GP does not have adhesive properties and may allow microfiltration at the gutta-percha-endodontic sealer (GP-CO) interface. Bioactive materials such as Bioglass 45S5 (BV) and copper-doped bioglass (BVCu) can contribute to antimicrobial action, bioactivity, chemical interaction and GP adhesion with bioceramic sealers. This study aims to evaluate BV application protocols in gutta-percha. The GP will be coated with BVCu through surface treatment (dip-coating), by immersion (IM) or atomization (AT) at concentrations of 1%. Gutta-percha blocks (n=11) will be made and distributed into the following experimental groups: G1: GP (control group); G2: GP AT BV; G3: GP IM VB; G4: GP AT BVCu; G5: GP IM BVCu. Specimens from different experimental groups will be coated with carbon and viewed under SEM at three magnifications (50x, 500x and 1000x) with or without immersion in PBS for 28 days. New GP blocks (n=78) will be made and distributed into experimental groups. Ready-to-use bioceramic endodontic cement discs Bio-C Sealer (Angelus, Brazil), BioRoot powder-liquid (Septodont, France) or AH Plus resin will be made with 10 mm in diameter and 5 mm in height. The GP blocks will be positioned on the filling sealer discs and stored for 48 hours in relative humidity. Subsequently, they will be coupled to the mechanical testing machine for tensile bond strength testing. The results obtained will be subjected to appropriate statistical tests with a significance level of 5%.
