Effect of the secretome of mesenchymal stem cells overexpressing BMP-9 and cultured on a microtopographic surface on bone repair

Abstract

The secretome present in the conditioned medium of mesenchymal stem cells (MSCs) genetically edited to overexpress bone morphogenetic protein 9 (BMP-9) (MSCsBMP-9) favors bone repair of mouse calvarial defects. As three-dimensionally printed surfaces on a microtopographic scale enhance the osteoblast differentiation of MSCs, we hypothesized that the secretome of MSCsBMP- 9 cultured on a surface with microtopography induces more bone repair than the secretome of MSCsBMP-9 cultured on a smooth surface. Thus, the aim of this study is to investigate the effect of the secretome present in the conditioned medium of MSCsBMP-9 cultured on a surface with microtopography on the bone repair of rat calvarial defects. The conditioned media of MSCsBMP-9 cultured on either smooth or microtopographic surface will be collected after 4 hours. Defects with 5 mm diameter will be created in rat calvarial grouped (n = 12 per group) according to the following treatments made with local injection of: (1) non-conditioned medium, (2) conditioned medium of MSCsBMP-9 cultured on smooth surface, and (3) conditioned medium of MSCsBMP-9 cultured on microtopographic surface. The treatments will be conducted 2 weeks post-defect creation and, 4 weeks after treatment, bone formation will be analyzed by microcomputed tomography and histological sections. The quantitative data will be submitted to the normal distribution test to determine the appropriate statistical test to be applied.
