Abstract

Orofacial pain, including those associated with Temporomandibular Disorders (TMDs), has a notable prevalence and complexity, with a multifactorial etiology. The therapeutic approach to TMDs requires a multidisciplinary perspective, including non-invasive therapies and, in some cases, surgical procedures. This study aims to evaluate the efficacy of a carbon monoxide donor (CORM-401) administered intra-articularly in attenuating persistent inflammation and pain induced by Complete Freund's Adjuvant (CFA) in rats. The animals will be distributed into six groups (n=9, each): Control (without CFA), Sham (surgical procedure without CFA) and Experimental (with CFA), administered with DPBS or CORM-401 vehicle. After seven days of induction of inflammation, the following evaluations will be carried out: 1) analysis of the response to mechanical allodynia in animals with persistent inflammation, compared to animals without inflammation (control); 2) measurement of temperature (classic sign of inflammation) in the temporomandibular joint (TMJ) region, in response to persistent orofacial inflammation; 3) quantification of the cytokines TNF-± and IL-1² in TMJ tissue. The results of this study have the potential to provide crucial insights for the development of more effective and personalized therapeutic approaches in the treatment of orofacial pain, with an emphasis on persistent inflammation.
