Development and analysis of hydraulic sealers to be used as repair materials in endodontics

Abstract

Endodontics involves the management of microbial infection. Its procedures encompass not only endodontic treatment, retreatment, and paraendodontic surgeries but also regenerative endodontics and vital pulp therapies. Hydraulic sealers are widely employed in all these areas of Endodontics due to their antimicrobial properties, sealing ability, and resistance, despite their handling difficulties and high solubility. This project will evaluate variations in the proportion of calcium silicate, radiopacifier, and additives in a ready-to-use experimental sealer to enhance its physicochemical and biological properties. Bismuth carbonate and zirconium oxide will be used as radiopacifiers, with calcium silicate proportions of 30%, 40%, and 50%. The liquid used will be propylene glycol containing 5% polyvinyl alcohol and 5% polyvinylpyrrolidone. The properties studied will include setting time, pH, radiopacity, flow, volumetric change, and solubility. Radiopacity, flow, and solubility tests will be evaluated according to ISO 6876/2001 standards. Setting time will be assessed according to ASTM C 266-2008 standard for hydraulic sealers. For pH and volumetric change tests, retrograde fillings will be performed, and volumetric loss will be analyzed by micro-computed tomography after 7 days of filling. The pH of the water in which the teeth are immersed will be measured with a pH meter at 3, 24, 72, and 168 hours. Experimental cements with promising results will undergo biological tests, including cell culture (cell viability test - MTT ASSAY). Data obtained from the tests will be statistically compared (significance level of 5%) with the results of Bio-C Repair and Sealer 26 thickened sealers.
