Abstract

This application for the Research Internship Abroad Scholarship (BEPE) is related to the doctoral project of the fellow Cristiane F. Benevente, titled: "Hypophysation of matrinxã, Brycon amazonicus, and lambari, Astyanax altiparanae, with lambari pituitary extract associated with PGF 2¿". The purpose of this BEPE application is to connect with the second objective of the student's thesis, which is: "to define and compare in vitro, using lambari follicles, the final maturation-inducing capacity (GVBD, 'germinal vesicle breakdown') and ovulation of lambari pituitary extract (EBHL), carp pituitary extract (EBHC), 17alpha, 20beta, dihydroxy-4-pregnen-3-one (DHP), and prostaglandin F2alpha (PGF2alpha)."The application focuses on the results obtained with lambari follicles, in which it was demonstrated that the application of 1¿g/mL of DHP, either alone or in combination with 50, 500, or 5000 ng/mL of PGF2alpha, is effective in inducing ovulation. However, surprisingly and unexpectedly, we observed that a concentration of 5000 ng/mL of PGF2alpha, both alone and combined with DHP, is sufficient to induce in vitro ovulation in lambari. This finding is significant because PGF2alpha is usually associated with inducing ovulation but not with migration and GVBD. This discovery leads us to apply for this BEPE to extend the evaluation of the mentioned experiment. We will use the material already collected to analyze the expression patterns of nuclear and membrane progestagen receptors. We will compare these patterns across different treatments, using various doses and combinations of DHP and PGF2alpha, to better understand the results obtained in ovulation induction.
