In vivo study of the effect of different ozone presentations on peri-implant repair in rats treated with zoledronic acid.

Abstract

Patients subjected to prolonged use or high doses of bisphosphonates, as well as antiangiogenics during the treatment of malignant diseases or osteoporosis are at risk of developing drug-induced osteonecrosis of the jaw (OMIM) after implant installation. Ozone therapy has a positive influence on peri-implant repair and can be an alternative for the prevention of OMIM after implant installation in at-risk patients. The objective of this project will be to evaluate the effects of ozone therapy, as a prevention of OMIM, on bone repair around implants installed immediately after extraction of the lower first molar in rats treated with zoledronic acid. Eighty adult male Wistar rats will be used, which will be treated with four applications of 0.035 mg/kg of zoledronic acid (ZOL) caudally with 15-day intervals between applications and will subsequently have implants installed immediately after extraction of the lower right molars. After the surgical procedure, the animals will continue treatment with ZOL in the same protocol until euthanasia. The animals will be divided into five groups (n=8) according to the proposed therapies: GS (Salt - ZOL will not be applied and ozone will not be administered); GC (Control), application of ZOL without ozone administration; GG (Ozone Gas), 60ug/ml of ozone gas will be applied to the peri-implant mucosa; GO (Ozonized oil) 0.1ml of oil will be applied to the peri-implant mucosa; GGO (Ozone Gas and Oil) will be applied 0.1ml of ozonized oil and 60ug/ml of ozone gas will be administered to the peri-implant mucosa. Therapies in the GG, GO and GGO groups will be carried out every 4 days, starting immediately after implant installation and remaining for 4 weeks. For in vivo analysis, in the mineralized section stage, 40 animals will be used, which will receive calcein and alizarin and 28 days after installation of the implants, they will be euthanized for evaluation through microtomographic analysis, confocal laser microscopy and histometric analysis. Forty animals will be used for biomechanical analysis and topographic characterization of the implant surface. Statistical analyzes will be carried out using a significance level of 5% (p<0.05).
