Effect of the application of copper-doped bioglass and tricalcium silicate in the treatment of dentin, gutta-percha and interaction with bioceramic sealers

Abstract

Gutta-percha (GP) associated with root canal sealer is the solid material used in root canal obturation. However, GP lacks adhesive properties, which may allow infiltration at the gutta-percha-sealer interface (GP-S). Furthermore, treating dentin with solutions containing biomaterials can enhance bioactivity and the interface between bioceramic material and dentin. Bioactive materials such as copper-doped bioactive glass (BVCu) and tricalcium silicate (ST) may contribute to bioactivity, antimicrobial action, chemical interaction, and adhesion of GP with bioceramic cements. This study aims to quantitatively and qualitatively assess the effect of dentin treatment, GP treatment with copper-doped bioactive glass (BVCu) solutions, and tricalcium silicate (ST) solutions. Subprojects 1 and 3: Aim to evaluate GP associated with BVCu and ST through surface coating (RS) or incorporation (IN). GP disks (n=18) with 10 mm diameter and 4 mm thickness will be prepared by: RS preparation: GP (control group); GP-BVCu 5%; GP-BVCu 10%; ST 5%; ST 10%. Surface analysis using Scanning Electron Microscopy (SEM) will be performed after immersion in PBS for 28 days. New disks will be prepared for tensile testing after contact with bioceramic cement Bio-C Sealer (BCS) and resin AH Plus (AHP): GP/BCS; GP-BVCu 5%/BCS; GP-BVCu 10%/BCS; GP/AHP; GP-BVCu 5%/AHP; GP-BVCu 10%/AHP; GP-ST 5%/BCS; GP-ST 10%/BCS; GP-ST 5%/AHP; GP-ST 10%/AHP. The tensile test will be conducted using a universal mechanical testing machine (Emic DL 2000). For IN preparation: GP disks with 10 mm diameter and 4 mm thickness will be prepared by IN: GP (control group); GP-BVCu 1%; GP-BVCu 2%; GP-ST 1%; GP-ST 2%. Subprojects 2 and 4: Aim to assess the effect of dentin treatment with BVCu and ST solutions on the dentin surface and bond strength with endodontic cements. Root canals of human teeth (n=20) will be prepared and analyzed according to dentin treatments using Scanning Electron Microscopy (SEM). Bovine teeth roots will be used to create circular disks of 20mm x 2mm (diameter x height), distributed according to dentin treatment: distilled water (DW), BVCu 5% solution; BVCu 10%; ST 5% solution; ST 10%, and subdivided according to materials: Bio-C Sealer (BCS, Angelus, Brazil) (n=24) or BioRoot RCS (BR, Septodont, France) (n=24). Mechanical tests will be performed using a mechanical testing machine (EMIC DL 2000). Data will be analyzed using analysis of variance (ANOVA) and Tukey's post hoc test with a significance level of 5% (p<0.05).
