Abstract Obesity is a critical risk factor for cognitive decline and neurodegenerative diseases, such as Alzheimer's, due to chronic inflammation and central insulin resistance. Jamelão (Syzygium cumini), a fruit native to Brazil rich in anthocyanins and flavonoids, emerges as a promising strategy for neuroprotection via the gut-brain axis. This project aims to evaluate the bioactive potential of jamelão in preventing obesity-associated cognitive impairment through an integrated multidisciplinary approach. The methodology includes chemical characterization of the fruit, in vitro digestion simulation (INFOGEST), and in silico molecular docking studies. The neuroprotective potential of bioaccessible fractions will be tested in a cell co-culture model (Caco-2/Astrocytes/Pericytes) to assess modulation of the NF-kappaB inflammatory pathway. Concurrently, in vivo assays with obese mice will investigate the effect of supplementation on spatial memory and systemic metabolism. Finally, advanced nutrigenomic analyses (BEPE/USA) will use the Affymetrix Clariom Assay platform to identify global gene expression and interaction networks of non-coding RNAs in brain tissue. We expect to elucidate the molecular mechanisms by which jamelão mitigates neuroinflammation, contributing to the valorization of national natural resources and the advancement of precision nutrition.
