Abstract

High mobility group box 1 (HMGB1) is a highly conserved protein present in the nuclei and cytoplasm of nearly all cell types. Recent investigations have indicated that HMGB1 has proinflammatory cytokine activities when released in the extracellular milieu. Thus, it has been suggested that HMGB1 is involved in the process of several acute and chronic inflammatory diseases. This secretion occurs by activated cells after inflammatory/infectious stimuli as well as by cells in death process. Moreover, the release depends on the extracellular stimuli as well as of the cellular type. In this way, activated macrofages and monocytes, neutrophils, mature dendritic cells and natural killer cells release actively HMGB1. In the extracellular milieu, HMGB1 acts through RAGES, TLR2 and TLR4 signaling. Thus, the mechanisms underlying HMGB1 expression and activity are complex and only partially understood and new studies using different cell types and in different pathological conditions can be useful. In this context, it has been demonstrated the activity of HMGB1 as a latter mediator in many acute and chronic inflammatory diseases, including sepsis, atherosclerosis and rheumatoid arthritis. Periodontal disease is a chronic inflammatory disease where the immune/inflammatory response has a fundamental role. Considering the similarities verified in the immunopathogenesis of periodontitis and other chronic/inflammatory diseases (including rheumatoid arthritis) an important role of HMGB1 can be suggested in periodontal disease process. Thus, the general purpose of the following study is to evaluate the expression pattern of HMGB1 in relevant periodontal tissue cells of murines.
