Effect of freeze-dried jaboticaba peel (Myrciaria sp.) in autophagy markers from adipose tissue in mice

Abstract

The obesity is characterized by excessive accumulation of white adipose tissue, with complex and multifactorial etiology. Among the disease¿s physiopathological mechanisms, the adipocyte hypertrophy and hyperplasia is known to trigger a chronic low-grade inflammation, with systemic consequences. These alterations, generated by the increase in cellular stress can be verified with the help of important molecular changes. One of the processes affected is the autophagy, a fundamental mechanism of cellular degradation important to the homeostasis and with different effects in each cell type. In adipose tissue, increased autophagy is related to adipocyte hypertrophy and hyperplasia, cellular stress and inflammation. In this way, the investigation of compounds that may contribute to the reduction in this process could be a strategy for obesity combat. In this way, the benefits of bioactive compounds consumption presents in food are inserted. Jaboticaba (Myrciaria sp.) is a native fruit from South America rich in phytochemicals in peel, whose consumption results in improved health effects, such as reduction of oxidative stress, lipid peroxidation and cardiovascular benefits. The objective of this study was to evaluate the effect of freeze-dried jaboticaba peel or its extract under the autophagy modulation in adipose tissue of C57BL/6 mice fed with high-fat or normocaloric diet. The supplementation was performed by two ways: through whole freeze-dried jaboticaba peel in diet, for 4 weeks, or through gavage of freeze-dried jaboticaba peel hydroethanolic extract (approximately 10mL/kg mice weight). The results showed that jaboticaba peel was effective in the reduction of autophagy signalization (reduction in the concentration of Beclin-1 and LCBII protein and increase in the concentration of SQSTM/p62 protein) in epididymal adipose tissue, in both forms of treatment, in animals fed with high-fat diet. However, the dose required for extract was lower than amount of jaboticaba peel ingested by supplementation, what do the extract consumption more potent. In addition to autophagy modulation in adipose tissue, jaboticaba peel extract improved the lipid profile and increased the insulin sensitivity in mice fed a high-fat diet (AU)
