Synthesis and characterization of poly (n-vinyl-2-pyrrolidone) hydrogels containing nano silver and laponite RD clay for modulation of neomycin release

Abstract

Current medical procedures increasingly require new biomaterials. In this context, hydrogels that are already widely used biomaterials become fundamental for health sectors. The objective of this work was to develop a hydrogel with characteristics of a dressing: biocompatibility, fluid absorption and antimicrobial activity. Hydrogels are materials composed of crosslinked polymer chains and water. There are several polymer crosslinking techniques that can be used for the production of hydrogels. In this work, gamma irradiation cross-linking was used which, in addition to forming covalent bonds between polymer chains, can sterilize the material making it ready for use in a single step. Two antimicrobial agents were used to obtain the hydrogels: neomycin and silver nanoparticles. Neomycin is an antibiotic widely used in wound care and silver nanoparticle is a historically known biocidal agent that has not yet shown any microbiological resistance. The silver nanoparticles of this work were produced via gamma irradiation and stabilized with laponite clay, a method that was very efficient in yield, stability and particle size. The influence of laponite clay on the physicochemical properties of gamma irradiation crosslinked PVP, PEG and agar was also studied, showing a significant influence of laponite on the reticulum structure and its characteristics. The hydrogels that contained neomycin and silver nanoparticles showed both bactericidal and fungicidal action showing that this material has a broad spectrum of action, besides no cytotoxicity. (AU)
