Resumo

Os microrganismos representam importante papel no estabelecimento das alterações endodônticas. Diferente da identificação fenotípica, o sequenciamento do 16S rRNA fornece identificação inquestionável até mesmo para bactérias que são raras. Isto toma possível identificar espécies que se acreditava não estarem sendo cultivadas, ou até encontrar espécies que nunca antes foram encontradas. O mesmo acontece quando se realiza a clonagem e sequenciamento do gene 16S, uma técnica que independe da cultura e capta também microrganismos não viáveis. O objetivo do presente estudo é identificar bactérias cultiváveis isoladas de canais radiculares infectados sintomáticos portadores de lesões periapicais, utilizando cultura e identificação por métodos bioquímicos e sequenciamento do gene 16S rRNA e identificar bactérias cultiváveis ou não das mesmas amostras por clonagem e sequenciamento do 16S rRNA. Serão coletadas amostras de 30 dentes com necessidade de intervenção endodôntica, com presença de lesão periapical e sintomatologia endodôntica, usando pontas de papel estéreis, transporte em VMGAIII. As amostras serão diluídas, plaqueadas e incubadas em anaerobiose. Colônias microbianas serão isoladas, caracterizadas e identificadas por métodos bioquímicos padronizados. Reação de PCR será realizada para amplificar a região V6-V8 do gene 16S rRNA e então será realizado o sequenciamento destas amostras. Além disso, 15 dessas amostras serão clonadas para sequenciamento do gene 16S rRNA. As sequências do 16S rRNA serão editadas e alinhadas usando o BioEdit e ClustalW. O BLASTn algorithm será usado para comparar as sequências de ácido nucleico contra seqüência de nucleotídeos do banco de dados do NCBI. Os dados serão tabulados e estatisticamente analisados pelo SPSS for Windows. (AU)
