Neurobiologia da doença de Alzheimer: marcadores de risco, prognóstico e resposta terapêutica

Resumo

Por meio de dois projetos temáticos apoiados pela FAPESP desde 1997, nosso laboratório desenvolve pesquisa translacional aplicada à análise de anormalidades do metabolismo de fosfolípides de membrana, sua influência na homeostase cerebral e suas implicações em doenças neuropsiquiátricas. No presente projeto, avaliaremos múltiplos mecanismos biológicos relacionados à fisiopatologia da Doença de Alzheimer (DA), buscando os correlatos clínicos que determinam um maior risco de demência em pacientes idosos com transtornos cognitivos e do humor. Este projeto Temático é dividido em dois módulos. No módulo 1 (Estudos Clínicos, Laboratoriais e de Neuroimagem), determinaremos de forma integrada os marcadores biológicos que influenciam o risco, o curso clínico e a resposta terapêutica; avaliaremos polimorfismos genéticos, biomarcadores em líquor e sangue periférico, e parâmetros de imagem cerebral estrutural e funcional (MRS e PET). Nosso maior interesse é estabelecer o diagnóstico precoce da DA e avaliar a eficácia de intervenções farmacológicas e não farmacológicas sobre a atenuação das perdas cognitivas, almejando a prevenção da demência. No módulo 2 (Neuroquímica e Biologia Molecular), utilizaremos modelos experimentais in vitro (culturas primárias de neurônios) e in vivo (camundongos transgênicos que acumulam peptídeo beta-amilóide, Aβ) para estudar os mecanismos moleculares que inter-relacionam diferentes processos patogênicos. Avaliaremos a toxicidade do Aβ e seu impacto sobre a neurogênese, a atividade enzimática e a expressão gênica. Em modelos pré-clínicos, investigaremos a possibilidade de modificar estes processos pelo tratamento com lítio, dadas suas propriedades neurotróficas e neuroprotetoras, integrando assim as vertentes experimentais e clínicas deste projeto.
