Resumo

Previously we have described the apoptosis occurrence when VERO cells are cultured under glutamine or galactose depletion. The aim of this paper is to extend the details of the main morphological features occurring in VERO cell lineage when cultured in nutrient depleted culture medium by electron and fluorescence microscopy and DNA laddering assays. The following changes were observed by light microscopy: brightness and progressive detachment of cells from monolayer, formation of cell surface in folds with multiple blebs and finally release of several membrane-bound apoptotic bodies. At ultra structural level, the apoptotic cells were characterized by condensed chromatin, intensive nuclear fragmentation, cytoplasmic organelle disruption, formation of apoptotic bodies and cell shrinkage. DNA electrophoresis in agarose gel of apoptotic VERO cells revealed an atypical pattern of bands, different from that usually observed for other cell lineages that can be used as a marker for VERO cell apoptosis.
