Abstract
The purpose of this Project is the identification of changes induced by hormones, drugs, ageing, expression of tumor suppre4ssor genes or genes involved in some pathologies, on DNA content, DNA methylation, and chromatin organization (supraorganization) and extensibility, as assessed by image analysis, cytochemistry, immunocytochemistry, and especial microscopies. Items to be investigated are as follows. 1) Action of estrogen and its metabolites, progestin, and anti-estrogenic drugs on chromatin of human breast epithelial cells (MCF-10F) and of tumors generated in SCID mice by injection of 17-beta-estradiol-transformed MCF-10F cells; 2) Chromatin organization remodeling as affected by lysyloxidases; and 3) Chromatin supraorganization and nuclear matrix glycoproteins in mouse hepatocytes following growth, aging and diabetes expression. The data so obtained will be correlated to cellular physiological changes. (AU)
